Gene expression levels constrain the feasible enzyme levels. Therefore, mRNA expression levels might be useful as constraints on flux levels in a constraint-based model. In this method, genes with lower expression levels will be used as a justification of giving a lower upper bound on the associated reaction fluxes. Mycobacter tuberculosis microarrays were used to constrain the model and then the theoretical maximal mycolic acid production yields were computed. Arrays for samples with know drugs which target mycolic acid production usually supported a decreased maximal flux through the mycolic acid biosynthesis pathway in the model.
